If you're not a "ferry person", or if you just want to get there as quickly as possible, a flight from Barnstable Municipal Airport in Hyannis will have you on-island in no time.
And you'll have a great view along the way!
Locally known as "AKC Air" (for Nantucket's airport code, ACK). Nantucket Air flies the Hyannis-Nantucket route daily, year round.

FYI: Cape Air airlines also offers flights between Martha's Vineyard and Nantucket. Check their site for details on the inter-island flights.
Flights between Hyannis and the islands are occasionally delayed or canceled on account of weather. So it's always good to have a travel "Plan B" in mind.
Plan B: A ferry ride. Unless Mother Nature is blowing up a gale, the Nantucket and Martha's Vineyard ferries will likely be making the trip across the Sound.
